Position Description
Gel-forming mucins are large and mosaic O-glycosylated molecules that constitute the framework of mucus hydrogels. The highly conserved CYS domain found in many mucins may crosslink mucin fibers to strengthen the mucin network and the mucosal barrier. Our aim is to show that recombinant molecules made of consecutive copies of the CYS domain secreted by a transgenic mouse line, by a mucus-secreting cell line and by a yeast strain reinforce the mucus barrier which will be microrheologically evaluated by analyzing particle dynamics (diffusion, sedimentation and/or confinement) and cell motility. The Proof of Concept that molecules made of CYS domains strengthen the mucus layer will be translated to an experimental mouse model of infection. We will demonstrate that recombinant molecules favor the mucus colonization with probiotic lactobacilli. Our results will empower to conceive scientific solutions in health for diseases linked to dysbiosis.
